Identification of Staphylococcus aureus on Length and Sequence Polymorphisms of 16S - 23S rRNA Spacer Region and the Comparison of Sequence Alignment


Abstract
Length polymorphisms in the PCR amplification product of 16S-23S spacer region rRNA in the 85 strains of Staphylococcus aureus and DNA sequence of 16S-23S spacer region rRNA of Staphylocorcus aureus CU 1103 were determined and compared with three other sequences of 16S-23S spacer region of methicillin resistant strain of Staphylococcus aureus by multiple alignment. Bands of twelve different length appeared with the range of the length of 160 to 1100 by predominant length ranged from 460 to 500 bp. Identity between DNA sequence of Staphylococcus aureus CU 1103 and three other sequences of gene bank showed hypervariability with the similarity of 43%-56%, and there were conserved loci of 120 by and 40 by at both ends of their sequences. Identification to the strain level seemed possible based on the length and sequence polymorphism of the 16S-23S spacer region in Staphylococcus aureus.
